Research

       The National Institutes of Health (NIH), Center for Disease Control (CDC), and foundational sources have supported our research, which addresses mechanisms controlling the maintenance of pregnancy and the endocrinology of the human maternal-fetoplacental unit.  Investigation of conditions regulating conceptus growth, pregnancy maintenance, and the initiation of normal labor is necessary to address the alarming rate of infant morbidity and mortality associated with human fetal growth restriction and prematurity. Therefore, we have utilized both in vivo (human, baboon, and rhesus monkey) and in vitro (human placental cell culture) paradigms, combined with both traditional and molecular methodologies, to elucidate mechanisms regulating the endocrinology of human pregnancy.  In this capacity, we have delineated estrogen’s role in maintaining progesterone synthesis via the low-density lipoprotein (LDL) receptor in the placental syncytiotrophoblast, as well as the potential for alternate cholesterol sources to fuel the progesterone production necessary to maintain threatened pregnancies. 

          An interest in fetal toxicology has resulted in identifying cadmium, an industrial pollutant and constituent of tobacco smoke, as an inhibitor of progesterone synthesis, thus linking premature labor with maternal exposure.  Another major interest in our laboratory involves leptin, a polypeptide hormone produced by fetal/maternal adipose tissues and the trophoblast that promotes conceptus development.  To this end, we have defined leptin/leptin receptor ontogeny with advancing primate pregnancy, proposed a role for placental estrogen in up-regulating the synthesis and release of leptin from maternal adipose tissue, and suggested a role for leptin in regulating fetal lung maturity.  Presently, collaborative work focuses on the effects of placental malarial infection on trophoblast endocrinology, pregnancy maintenance and the preservation of the maternal-fetal interface as an immunologically privileged site.  In addition, collaborations with others currently involve our interaction on projects examining the effects of West Nile Virus on placental function, conceptus development, and premature labor.
